I imagine he was pretty fucked up to begin with to be able to play the Joker to this level
like Hugh Laurie doing Dr. House, he's a method actor too, but there are a lot of traits House has that are simply Hugh's traits amplified, the only thing he says he really has to work on is the limp and the "American" accent (Hugh has a VERY thick British accent, especially noticable by anyone who watched Black Adder)
House's "deep moody depression" is actually just an amplified version of Hugh's own clinical depression, something he's been seeing a therapist about since the mid 90's
So I really think most method actors simply work on amplifying their own preexisting traits.
but this is one of those nature of nurture arguments, was he crazy before hand (which I believe so), or did this character put him over the edge?
this is still assuming his OD was intentional, I've known more than a few people who have gotten seriously loaded on things so much so that they don't think that taking something else could put them under, next thing you know I'm sitting in an emergency room waiting room while they get their stomach pumped, but that was in my rowdy college days
Either way, it should be an outstanding movie and I will be seeing it sometime next week after the weekend rush is over but while its still in the "big" theater rooms.
Very strange for me to say since I actually hate Ledger as an actor, but from what I hear and seen so far he's done a great job.

And yeah, Aaron Eckhart played Harvey Dent flawlessly. I've only seen him in Thank You for Smoking, so this was a pretty good look at his range. For being a theatrical release, it's among the darkest movies I've ever seen, especially considering it's not a horror or psychological thriller.
